Membránový transport

Membránový transport Pozorování transportéru sodných a draselných kationtů v buňkách kvasinek pomocí fluorescenčního mikroskopu. Zeleně je označen protein nacházející se v plasmatické membráně buněk, červeně je označena membrána vakuoly.

Studujeme proteiny transportující látky a signály přes buněčné membrány.  Tyto proteiny, nazývané transportéry, zajišťují přísun živin do buněk, odstraňování odpadních látek z buněk a také komunikaci s okolním prostředím.  Při studiu transportérů živočišných a rostlinných buněk využíváme převážně modelový eukaryontní mikroorganismus – kvasinky. Zabýváme se výzkumem v těchto oblastech:

  • Struktura, funkce a regulace buněčných transportních systémů na úrovni molekul proteinů
  • Úloha transportérů ve specifických vlastnostech buněk a při vzniku onemocnění
  • Úloha transportérů v pathogenicitě a virulenci kvasinek rodu Candida
  • Vývoj metodik pro stanovení fyziologických parametrů na buněčné úrovni

Homeostáze kationtů alkalických kovů a pH v buňkách

Koncentrace draselných a sodných iontů, jakož i koncentrace protonů (pH), je v buňkách přísně regulována na úrovni řady membránových proteinů, které přes buněčné membrány přenášejí kationty a protony pomocí různých transportních mechanismů. Nesprávné fungování některých transportérů může mít za následek vážné poruchy a onemocnění. Detailně proto zkoumáme úlohu jednotlivých transportérů a vliv jejich aktivity na zdraví buněk.  Více

Úloha transportérů ve specifických vlastnostech nekonvenčních kvasinek

Některé, tzv. nekonvenční druhy kvasinek přežívají velké výkyvy okolního pH, teploty, koncentrace živin a iontů nebo osmotického tlaku. Snažíme se identifikovat a charakterizovat specifické transportéry, které se svojí aktivitou podílejí na schopnosti těchto druhů přežít různé extrémní podmínky. Získané znalosti také mohou napomoci ve zlepšování vlastností průmyslově používaných kvasinek. Více

Naše oddělení se dlouhodobě zapojuje do projektu Otevřená věda, který je organizován Akademií věd ČR, v.v.i. a má za úkol přilákat na vědecká pracoviště nadějné studenty středních škol.
